ABSTRACT:
The research was presented in two parts. Part-I deals with the synthesis of Tolmetin sodium, an effective anti-inflammatory agent for the treatment of arthritis and other inflammatory disorders. In this we have developed a simple and convenient laboratory scale method for synthesis of the aforementioned drug. This method involves the use of readily available and inexpensive chemicals, with quantitative yield of the final product on large scale too.

Part-II concerns with QSAR analysis of PDE4 inhibitors as antiinflammatory agents. In the present study, two QSAR methods namely, Multiple Linear Regression (MLR) Analysis and Genetic function approximation (GFA) were employed to gain insights into the factors influencing PDE4 enzyme inhibition and to develop predictive model for the design of NCEs as PDE4 inhibitors with antiinflammatory activity. The former method yielded statistically insignificant models. In order to get statistically significant models robust method, GFA was employed. The models generated by GFA are consistent with statistical significance. The study hints that shape, structural, and electronic parameters are influencing the biological activity. The generated models can be helpful in the design of novel, selective PDE4 inhibitors as potent antiinflammatory agents.

ABSTRACT:

Heterocyclic systems are one of the most important classes of organic compounds present in nature or synthesized in the laboratory. These compounds possess an array of biological activities and are employed in the treatment of commonly occuring diseases.

Literature survey revealed that Chalcones, Pyrimidines and Pyrazolines possess a broad spectrum of biological activities like antimicrobial, anti-inflammatory, antimalarial, antidepressant, anti- histamine, antitubercular and anticancer. This thesis is an endeavour in this direction of synthesis and characterization of such compounds based on Elemental analysis, IR, 1H NMR and Mass spectroscopy. The pharmacological and microbial screening of synthesized compounds has also been included.

Chalcones afford a facile route of access to many heterocyclic systems containing Nitrogen and Oxygen. Hence an attempt is made to synthesize chalcones from 3'- methyl - 4'- hydroxyacetophenone with various substituted aromatic / heterocyclic aldehydes by the Claisen – Schmidt condensation. The resulting chalcones after purification and characterization by physical and spectral methods have been converted successfully into Pyrimidines and 2 – Pyrazolines by reaction with Guanidine hydrochloride and Phenylhydrazine hydrochloride respectively. The resultant compounds were identified by physical and spectral methods and were also screened for selected biological activities based on the reported literature. These efforts resulted in the synthesis of some promising compounds with significant biological activities.
